Novel factors and functions at the interface of transcription and mRNA export

Histone modifications, gene transcription, mRNA biogenesis and export of transcripts orchestrate gene expression in eukaryotic cells. A cascade of cotranscriptional chromatin modification events, including monoubiquitination of histone H2B (H2Bub1) by Rad6 and Set1-dependent histone H3 methylation at lysine 4 (H3K4me), ensures the proper metabolism of the transcripts that have to be exported to the cytoplasm. However, a complete description of the coordination of the different steps is still incomplete. Here I will present part of our new data showing that, Sus1, which is the shared component of the SAGA deubiquitination module and the Transcription/Export complex (TREX2), associates broadly to most yeast genes. Notably, heat shock drastically affects Sus1’s binding pattern. Moreover, we demonstrate by genomic-run-on experiments that Sus1 is required for both synthesis and stability for a wide range of transcripts. Our study provides support for the new model for SAGA in transcription acting as a global transcriptional regulator. In addition, I will also present recent work demonstrating a novel role for the Ran-binding protein, Mog1 in gene expression.
